Affini-T Therapeutics announced on March 22, 2022, that it had completed an oversubscribed $175 million financing co-led by Vida Ventures and Leaps by Bayer. The Fred Hutchinson Cancer Research Center co-founded company said it would use the proceeds to advance engineered T-cell therapies aimed at solid tumors. The financing was a 2022 announcement, not a current fundraising update.

What is Affini-T Therapeutics?

Affini-T Therapeutics is a biotechnology company developing engineered T-cell therapies for cancers with specific genetic drivers. In its financing announcement, the company described a platform combining T-cell receptor (TCR) discovery with synthetic biology components. Its stated targets included mutations in KRAS and p53, with the aim of developing treatments for solid tumors. These were development goals, not evidence that the therapies had demonstrated clinical benefit.

The company’s announcement, published by Bayer in 2022, said KRAS mutations account for up to 30% of all cancers. That is Affini-T’s attributed figure; the announcement did not identify its original epidemiological source, so it should not be read as an independently verified estimate. The company highlighted lung, colorectal, and pancreatic cancers as settings where KRAS mutations are especially frequent.

Affini-T said it planned to use the proceeds to operationalize its platform discovery engine, move multiple programs targeting oncogenic drivers toward clinical development, and pursue complementary technology licenses. These were the company’s intended uses of the funds, rather than guarantees about later development milestones.

What is Affini-T developing for solid tumors?

The company’s approach centers on TCR-T cell therapy: engineering a patient’s T cells to recognize cancer-associated targets, including targets linked to oncogenic mutations. Affini-T described combining receptor discovery with synthetic biology to build its platform. CEO and co-founder Jak Knowles characterized the strategy this way: “By targeting oncogenic drivers like mutated KRAS, we strike at the core genetic mutations that enable tumors to grow and spread.” This is the company leader’s description of the rationale, not proof of an established treatment effect.

The company’s later pipeline information provides a dated snapshot, not a current status report. A non-confidential Affini-T presentation from March 2025 listed AFNT-111 and AFNT-211 in Phase 1 and showed an IND clearance milestone for AFNT-212 planned for the first half of 2025. A planned milestone is not confirmation that it occurred, and those deck entries do not establish present trial status or results.

What is Affini-T’s connection to Fred Hutch?

Affini-T described itself as co-founded at Fred Hutchinson Cancer Research Center and identified Phil Greenberg, Aude Chapuis, and Tom Schmitt as scientific co-founders. Fred Hutch was also listed as an investor in the 2022 financing. Greenberg, whom the announcement identified as leader of Fred Hutch’s Program in Immunology and holder of the Rona Jaffe Foundation Endowed Chair, said: “Cell therapies have revolutionized the treatment of cancer, but challenges remain in making this powerful technology effective for a greater variety of cancers.”
