Short answer: Retro Biosciences was reported on January 24, 2025, to be seeking up to $1 billion for therapies intended to add 10 healthy years to human life. That was a fundraising target, not proof that the company had extended anyone’s lifespan. On May 22, 2026, Retro announced an initial financing close at a $1.8 billion pre-money valuation, but did not disclose how much cash it had raised or confirm that the full $1 billion target was closed.
What the January 2025 announcement actually said
TechCrunch, Endpoints and Fortune reported that Retro Biosciences was pursuing a financing round of up to $1 billion. The proposed capital was intended to advance several drug programs toward human testing, alongside work in cell therapy, tissue reprogramming and protein engineering. The company’s stated ambition is to add 10 healthy years to human lifespan, a mission rather than a clinically demonstrated result.
The wording matters: “raising $1 billion” described an in-progress financing effort. It did not establish that investors had already transferred $1 billion to Retro. Contemporary coverage included TechCrunch’s report, an Endpoints account and Fortune’s coverage.
The latest financing status
Retro’s May 22, 2026 announcement says it reached the initial close of a new financing round at a $1.8 billion pre-money valuation, led by 4P Capital and other investors. A pre-money valuation is the company’s agreed value before new investment is added; it is not the amount of money raised.

Because Retro did not publish the dollar amount in that initial close, the announcement does not prove that the company raised $1 billion. “Initial close” can indicate that a financing has additional closings, but Retro’s statement does not specify the final size or completion date. The company’s update is available at Retro’s financing announcement.

Sam Altman’s role—and what it does not mean
MIT Technology Review reported that Altman personally invested $180 million in Retro’s earlier financing. That figure refers to the company’s previous funding, not automatically to the 2025 target. “Backed by Sam Altman” therefore means personal financial backing, not that OpenAI owns Retro or supplied the reported $1 billion.
OpenAI and Retro have also collaborated on research. That relationship is separate from Altman’s personal investment, and Retro remains an independent biotechnology company. The earlier investment was described by MIT Technology Review.

What Retro is developing
Retro describes a portfolio rather than a single “anti-aging pill.” Its public pipeline covers conventional drug development and biological-engineering approaches aimed at age-related disease. These include:
- small-molecule and other drug programs;
- cell-therapy research and manufacturing;
- tissue reprogramming;
- AI-enabled protein engineering; and
- interventions aimed at cellular mechanisms associated with aging.
Its pipeline page and company materials present these as development programs. A mission to increase healthy lifespan does not necessarily become a regulatory indication called “aging”; medicines generally enter trials for specific diseases, symptoms or measurable biological conditions.
Where RTR242 stands
Retro’s 2026 update identifies RTR242 as its lead clinical candidate. The company says the program moved from indication selection to first-in-human dosing in 15 months, entered the clinic in 2025 and was in Phase 1 by May 2026. It also described additional first-in-human milestones planned for 2026 and 2027.

Third-party descriptions characterize RTR242 as a small molecule intended to restore lysosomal function, part of the cell’s recycling and autophagy system. That mechanism is reported, not proof of clinical benefit; the available public material does not establish that RTR242 extends life or treats a disease effectively.
Phase 1 is primarily an initial evaluation of safety, tolerability, dosing and sometimes biological signals. Even a successful Phase 1 study would not demonstrate longer human lifespan. Later controlled trials would need to show benefits for a defined disease or health outcome, with long-term evidence required for any lifespan claim.
How artificial intelligence fits in
OpenAI and Retro reportedly worked on GPT-4b micro, a model specialized for protein engineering and longevity research, as described by MIT Technology Review. A system like this may help generate protein designs, rank candidates or suggest biological hypotheses.

AI does not remove the need for laboratory experiments, toxicology, manufacturing controls or clinical trials. A model’s ability to propose a protein is not evidence that a resulting therapy works in people, and GPT-4b micro is not a consumer longevity treatment or a feature of ChatGPT.

Healthspan is not the same as lifespan
Retro’s wording focuses on healthy years—healthspan—rather than simply postponing death. A treatment could improve a particular age-related disease without extending total lifespan, or extend survival while leaving quality of life unchanged.
Aging has many biological causes
Aging involves interacting changes across cells, tissues and organs. A portfolio can spread risk across mechanisms, but it also means no single early result validates the company’s overall mission.

Clinical endpoints take time
Regulators generally evaluate medicines against defined diseases or measurable conditions. Lifespan is a long-term endpoint, so practical development is more likely to begin with safety, biomarkers and disease-specific outcomes than with a trial designed to prove an extra decade of life.
Early safety can outweigh ambitious claims
Cellular reprogramming, genetic manipulation and other rejuvenation strategies require particular attention to unintended biological effects. Until controlled human data are disclosed, claims about reversing aging or making people live longer remain unverified.
How to evaluate Retro’s progress
- Separate target from close. A reported $1 billion target is not the same as cash raised; the 2026 $1.8 billion figure is a valuation.
- Check the development stage. RTR242 being in Phase 1 means early human testing, not demonstrated efficacy.
- Look for the indication. Ask which disease or measurable condition each candidate is intended to treat.
- Prioritize independent evidence. Trial registrations, peer-reviewed results, regulatory filings and disclosed clinical data are more informative about efficacy than company financing announcements.
- Read AI claims narrowly. Discovery assistance can accelerate candidate selection but cannot substitute for validation in cells, animals and people.
Retro in the longevity-investing landscape
Retro is part of a broader group of heavily funded longevity companies, including Altos Labs and Unity Biotechnology, pursuing cellular reprogramming, regenerative medicine, senolytics and other approaches. Large capital reserves can support multiple expensive programs and manufacturing work, but portfolio breadth can make it harder to determine which mechanism will succeed.
For investors, the central question is not whether billionaires can finance longevity research. It is whether a specific candidate can produce a reproducible, clinically meaningful result for a defined disease or health outcome. A company can be scientifically ambitious and financially well supported while remaining years—or decades—from proving a lifespan benefit.
